There are a few landing strips up there, however I dont know what it takes to use them(permission). There is one in coldfoot,one at deitrich camp,and one at Galbreath lake. As for taking off on the road it could be done but you will have to decide where you are going and where or if you can land. The west side of the road somewhat close to the road is the park and is off limits for hunting but the east side is wide open. There are a lot of guides taking people out there but I dont know how you would find out where they are at. I know Chandalar lake has a lodge and hunting outfits going out of there and they land right on the lake. I wish I could be more specific but I can only imagine the hunting would be awesome if you could get 10-20 miles off the road where no one else is at as most of the guides are out farther than that.I have a friend that has been intersted in doing a trip like the one you describe in an ultralight but even with his experience flying an ultralight the winds are so unpredicatable up there, Supercubs crash every year I know of 2 that crashed or had damage upon landing this past August alone. Its a pretty expensive tow bill out of there if you know what I mean. A friend of mine was flown in on a supercub and the cub got damaged on landing had to wait days to be taken out and they had to come back and strip the cub and take what they could out one piece at a time, not fun.
